=== Substrate concentrations === The remaining enzymes of the cycle are controlled by the concentrations of their substrates. Thus, inherited deficiencies in cycle enzymes other than ARG1 do not result in significant decreases in urea production (if any cycle enzyme is entirely missing, death occurs shortly after birth). Rather, the deficient enzyme's substrate builds up, increasing the rate of the deficient reaction to normal. The anomalous substrate buildup is not without cost, however. The substrate concentrations become elevated all the way back up the cycle to NH+4, resulting in hyperammonemia (elevated [NH+4]P). === United States === US General Douglas MacArthur pointed out that before World War II, the US protected its western shores with a line of defense from Hawaii, Guam, to the Philippines. However, this line of defense was attacked by Japan with the Pearl Harbor bombing of 1941, thereby drawing the US into the war. The US subsequently launched the air Raid on Taipei (called Taihoku under Japan's empire) and launched the atomic bombings of Hiroshima and Nagasaki. The WW2 victory allowed the US to expand its line of defense further west to the coast of Asia, and thus the US controlled the first island chain. Between the end of WW2 and the Korean War, MacArthur praised Taiwan, located at the midpoint of the first island chain, as an 'unsinkable aircraft carrier'. In April 2014, the United States Naval Institute (USNI) assessed that the first island chain is the most effective point to counter any Chinese invasion. The US could not only cut off the People's Liberation Army Navy from entering the western Pacific, but also predict where they may move before trying to break through in the first place. The US and first island chain countries are able to coordinate because of the US military's freedom of navigation in the first chain block. A June 2019 article published by the United States Naval Institute (USNI) called for the navy to establish and maintain a blockade around the first island chain if the US was go to war with China.
=== Charles W. Post === The second major innovator in the cereal industry was Charles W. Post, a salesman who was admitted to Kellogg's sanitarium as a patient in the late 1800s. While there, he grew deeply impressed with their all-grain diet. Upon his release, he began experimenting with grain products, beginning with an all-grain coffee substitute called Postum. In 1897 (or 1898) he introduced Grape-nuts, the concentrated cereal with a nutty flavor (containing neither grapes nor nuts). Good business sense, determination, and powerful advertising produced a multimillion-dollar fortune for Post in a few years. After his death, his company acquired the Jell-O company in 1925, Baker's Chocolate in 1927, Maxwell House coffee in 1928, and Birdseye frozen foods in 1929. In 1929, the company changed its name to General Foods. In 1985, Philip Morris Tobacco Company bought General Foods for $5.6 billion (equivalent to $16.8 billion today) and merged it with its Kraft division. === Identifying Translated mRNA Regions === By using specific drugs, ribosome profiling can identify initiating regions of mRNA, elongating regions, and areas of translation stalling. Initiating regions can be detected by adding harringtonine or lactidomycin to prevent any further initiation. This allows the starting codon of the mRNAs throughout the cell lysate to be analyzed, which has been used to determine non-AUG sequences that do initiate translation. The other elongating regions can be detected by adding antibiotics like cycloheximide that inhibit translocation, chloramphenicol that inhibits transfer of peptides within the ribosome, or non-drug means like thermal freezing. These elongation freezing methods allow for the kinetics of translation to be analyzed. Since multiple ribosomes can translate a single mRNA molecule to speed up the translation process, RiboSeq demonstrates the protein coding regions within the mRNA and how quickly this is done depending on the mRNA being sequenced. This also allows for ribosome profiling to show pause sites within the transcriptome at specific codons.
