Early work on thymic extracts in the 1960s described a heat-stable acidic fraction containing many polypeptides. Separation of that mixture yielded individual components, and thymosin alpha-1 was named as one of them on the basis of assays for T-cell activity. The first preparations came from calf thymus, while subsequent research and clinical material has been chemically synthesized. Nomenclature in older papers is inconsistent, and the same peptide sometimes appears under different designations, which complicates literature searches.
Most published studies on thymosin alpha-1 report changes in immune measurements rather than clinical outcomes, and findings differ across designs and populations. Whether the peptide signals through one defined receptor or through several less specific interactions remains an open question. Its reported circulation half-life of a few hours complicates comparison of dosing schedules across trials. Mechanistic claims are frequently drawn from isolated cell cultures, and how far those results extend to whole organisms is unresolved.
Thymosin alpha-1 is a synthetic peptide of 28 amino acids whose sequence matches the amino-terminal region of prothymosin alpha. The chain is acetylated at its first residue and contains one disulfide bridge between two cysteine residues, which folds the molecule into a compact loop. Its molecular formula, C129H215N33O55, corresponds to a monoisotopic mass of roughly 3,106 daltons. Material used in laboratories is made by solid-phase synthesis rather than isolated from animal tissue.
Lyophilized material is typically treated as a hygroscopic solid that should be brought to room temperature before the container is opened, which limits condensation on the powder. Reconstitution is commonly done with sterile water or a buffered diluent, and gentle mixing is preferred over vigorous agitation to reduce foaming and surface adsorption. Because peptides can bind to plastic and glass, diluents containing a small amount of carrier protein are sometimes used in laboratory work. Working solutions are generally aliquoted and prepared fresh rather than subjected to repeated freezing and thawing.
Stability depends on temperature, pH, and the number of freeze-thaw events the sample has experienced. Freeze-dried material is commonly held at -20 °C or colder, while reconstituted liquid is kept cold and used within a short window. Extreme pH and prolonged light exposure can promote deamidation, oxidation, or aggregation, particularly at asparagine and methionine positions. Adsorption to container walls can lower the measured concentration of a dilute solution even when the peptide molecules themselves remain intact.
Identity and purity are usually assessed by reversed-phase high-performance liquid chromatography, which separates the target peptide from truncated or chemically modified byproducts. Mass spectrometry confirms the expected molecular mass and can indicate acetylation state or sequence errors. Amino acid analysis and peptide mapping supply complementary sequence-level information, while endotoxin testing is relevant for preparations intended for cell or animal work. Purity figures reported by suppliers refer to the method used and are not directly comparable across laboratories unless conditions are stated.

Immune signaling studies link thymosin alpha 1 to Toll-like receptor pathways, particularly TLR2 and TLR9, on dendritic cells and other antigen-presenting cells. Activation of these receptors promotes maturation of T cells and increases natural killer cell activity. The peptide shifts cytokine output toward a T helper 1 profile, raising interferon gamma and interleukin 2 while modulating interleukin 10. Whether these effects translate into clinical benefit for any specific disease remains a subject of debate. Reported outcomes vary across trials and populations.
Thymosin alpha 1 is approved as a medicine in several countries, including Italy and China, for indications such as chronic hepatitis B and as an immune adjuvant. It is not approved by the United States Food and Drug Administration as a therapeutic product. In research settings the peptide appears in studies of sepsis, vaccine response, and oncology support, often with mixed or inconclusive results. The evidence base is uneven, and reviews note that many trials were small. Regulatory status therefore differs widely between jurisdictions.
Thymosin beta-4 is a separate 43-residue peptide that binds actin and participates in cell migration; it shares no sequence similarity with thymosin alpha-1 despite the common family name. Other preparative materials, such as thymosin fraction 5 and thymopoietin, contain distinct mixtures or peptides. The shared thymosin label reflects the tissue of origin used in early purification, not a common structural core. Treating these molecules as interchangeable is a frequent source of confusion in laboratory reports and in popular summaries alike.
Thymosin alpha-1 is a synthetic peptide of 28 amino acid residues that corresponds to a naturally occurring fragment first isolated from thymus tissue. Its chain is acetylated at the amino terminus, a modification that shields the peptide from rapid cleavage by aminopeptidases. The molecule carries a net negative charge at physiological pH and dissolves freely in water. Researchers classify it as an immune-modulating agent rather than a classical hormone, because it acts on several cell types of both the innate and the adaptive immune system.
The peptide was identified during work in the 1970s on thymosin fraction 5, a partially purified extract of calf thymus. Investigators separated that mixture and characterized individual components, one of which they named thymosin alpha-1. The same compound later received the international nonproprietary name thymalfasin. Commercial material is produced by solid-phase peptide synthesis rather than by extraction, so synthetic and natural forms share an identical sequence. Naming conventions vary across the literature, and readers should distinguish the alpha-1 peptide from other thymosins that have unrelated sequences and functions.

== Inventions == James Watt did not invent the steam engine, nor were his ideas on steam engine power inspired by a kettle lid pressured open by steam. Watt improved upon the already commercially successful Newcomen atmospheric engine (invented in 1712 by Thomas Newcomen) in the 1760s and 1770s, making certain improvements critical to its future usage; his new steam engine later gained huge fame as a result. Although the guillotine was named after the French physician Joseph-Ignace Guillotin, he neither invented nor was executed with this device. He died peacefully in his own bed in 1814. Rather, it was Guillotin's speech favoring beheadings over other forms of execution that led to the device being referred to as "La machine Guillotine" and later simply guillotine. Thomas Crapper did not invent the flush toilet. A forerunner of the modern toilet was invented by the Elizabethan courtier Sir John Harington in the 16th century, and in 1775 the Scottish mechanic Alexander Cumming developed and patented a design for a toilet with an S-trap and flushing mechanism. Crapper, however, did much to increase the popularity of the flush toilet and introduced several innovations in the late 19th century, holding nine patents, including one for the floating ballcock. Thomas Edison did not invent the light bulb.
== Society and culture == In some parts of Africa, including Somalia, Ethiopia and Eritrea, the uvula or a section of it is ritually removed by a traditional healer. In this case, the uvula may be noticeably shortened. It is not thought to contribute to velopharyngeal inadequacy, except in cases where the tonsils have also been removed.

==== Anti-infective ==== Arsenic trioxide has shown efficacy against a range of pathogens due to its broad-spectrum antimicrobial, antiviral, and antiparasitic properties. However the significant toxicity of arsenic trioxide has greatly limited its use as an anti-infective agent. Historically, it was used to treat infections such as syphilis and trypanosomiasis before the advent of modern antibiotics. Recent research highlights its potential in combating multidrug-resistant bacteria, with certain sulfur-containing polyarsenicals exhibiting potent activity against Staphylococcus aureus, a major human pathogen. Arsenic trioxide has also demonstrated antiviral activity, notably inhibiting hepatitis C virus (HCV) replication at submicromolar concentrations and disrupting adenoviral infection by modulating host cell nuclear structures. The discovery of organoarsenical antibiotics like arsinothricin underscores arsenic trioxide’s potential as a direct antimicrobial agent targeting emerging and resistant pathogens.
The middle row was almost entirely damaged, although scenes of the Preaching Buddha were identifiable. The panels can be numbered 5 to 9, but 5 and 9 being half-panels going over the adjacent walls. In one of the panels appear soldiers similar to those of the Cave of the Painters. The bottom of the wall contained fragments of panels showing: 10) the Parinirvana, 11) devotees looking at the Buddha being put in a coffin, 11) The Buddha in his coffin, and 12) would have been the Cremation of the Buddha. The upper part of the mural was removed by Grünwedel, and sent to Germany in panels, where some are still held in the Museum für Indische Kunst. This presentation of the various events of the life of the Buddha in successive panels reminds of examples from Gandhara, such as the Sikri stupa, although the panels in the Cave of the Peacock are remarkable by their rigorous chronological arrangement. Similar types of narrative panels have also been found in Andhra. The dome over the cella is composed of eight pairs of segments filled with a flying apsara among peacock feather. Numerous devatas and Buddhas of the past are painted around the dome. According to Historian of Art Benjamin Rowland, commenting one of the remaining fragments, the "group of sword-bearing figures are recognizable Indian ethnic types". Pictures of monks and one Kuchean donor holding a basket of flowers, all labeled with Brahmi inscriptions, appeared on the door wall. In the art of Kizil explanatory labels were often added to pictures of donors.
AOD9604 is an orally active, lipolytic 16-amino acid peptide fragment of human growth hormone and derivative of the C-terminal domain of human growth hormone (HGH). It consists of HGH residues 176–191, with a tyrosine in place of the phenylalanine at the N-terminal end. Initial human trials showed that it retains the lipolytic properties of human growth hormone without stimulating IGF-1 production. However, later studies failed to demonstrate a sufficient lipolytic effect. Its development was terminated in 2007. Despite its limited efficacy, AOD9604 is still banned in athletes and tested for in competition. AOD9604 appears to enhance lipolysis in mice by upregulating beta-3 adrenergic receptors. Beta-3 adrenergic receptor knockout mice are unresponsive to the lipolytic effects of AOD9604. In a 12 week randomised clinical trial, subjects receiving AOD9604 lost, on average, 1.8 kg more than those receiving placebo. Development of AOD9604 was halted following poor efficacy in a later 24 week trial.

Its sequence corresponds to the amino-terminal portion of prothymosin alpha, a larger protein present in many cell types. The isolated 28-residue peptide is a fragment of that protein rather than a separately encoded molecule, and laboratory material is produced by synthesis.
The single bridge between two cysteine residues holds the chain in a folded loop that influences its shape and its behavior in solution. Loss of the bridge through reduction or oxidation shifts chromatographic retention and is tracked during stability work.
It is a defined 28-residue sequence derived from a larger precursor, whereas many other thymic preparations are mixtures of several polypeptides. Its acetylated amino terminus and single disulfide bridge distinguish it chemically from unrelated thymic extracts.
Cold storage slows the chemical degradation reactions that occur in solution. Lyophilized powder is more stable than reconstituted liquid and tolerates longer storage periods. Repeated temperature cycling should still be avoided because it can drive aggregation and loss of material.
