A practical reference on clinical endpoint: what it is, how it behaves, what the literature reports, and where the honest uncertainties sit.

Overall evidence quality varies considerably. A large share of published reports come from single centers, rely on surrogate immunological markers, or lack adequate control groups. Systematic reviews have highlighted this heterogeneity as a barrier to pooling results. Open questions include which patients, if any, might benefit, what treatment duration is appropriate, and whether any effect is independent of standard care. The peptide is often described as an immune modulator rather than a therapy for one disease, which complicates confirmatory trial design.
Thymosin alpha 1 was identified in 1977 as a component of thymosin fraction 5, a heterogeneous preparation used in early studies of thymic function. Investigators purified the active material and determined its amino acid sequence, which enabled chemical synthesis. Work in the following decades concentrated on T-cell maturation and immune reconstitution in animals and small human cohorts. Early preparations varied in composition, so results from that period are difficult to compare with studies using defined synthetic peptide.
Clinical research has examined the peptide in chronic hepatitis B and C, as a vaccine adjuvant, and in sepsis and oncology settings. Findings across trials are mixed; some report changes in selected immune markers, while others find no clear clinical benefit. Many studies are small and define outcomes differently, which limits comparison. Regulatory approval is confined to a few countries, and the compound is not an approved drug in the United States or most of Europe.
Identity and purity testing for thymosin alpha 1 relies mainly on reversed-phase high-performance liquid chromatography and mass spectrometry. Chromatography separates the parent peptide from truncated or modified variants, while mass spectrometry confirms the expected molecular mass. Amino acid analysis and peptide mapping provide additional sequence confirmation. Counterion content, water content, and residual solvents are measured separately as part of specification testing. No single method captures every attribute, so laboratories combine several techniques.
The peptide lacks cysteine, methionine, and tryptophan, so disulfide scrambling and sulfur oxidation are not major degradation routes. Instead, aspartate residues can undergo isomerization or cyclization to succinimide intermediates, generating isoaspartate variants. Hydrolysis of peptide bonds also occurs slowly in solution. These changes may reduce biological activity even when the main peak remains detectable. Stability studies therefore track both potency and the appearance of related substances.
Lyophilized thymosin alpha 1 is typically stored refrigerated at 2 to 8 degrees Celsius and kept away from light. Reconstituted solutions are less stable and are usually used promptly after preparation. Repeated freeze-thaw cycles are avoided because they can promote aggregation and loss of activity. The peptide adsorbs to some plastic and glass surfaces, so a carrier protein is often added to dilute working solutions. Manufacturer instructions and published protocols both govern handling.

=== Spondylodysplastic === Spondylodysplastic EDS (spEDS) is characterized by short stature (progressive in childhood), muscle hypotonia (ranging from severe congenital to mild later-onset), and bowing of limbs. It can be caused by variations in both copies of the B4GALT7 gene. Other cases can be caused by variations in the B3GALT6 gene. People with variations in this gene can have kyphoscoliosis, tapered fingers, osteoporosis, aortic aneurysms, and problems with the lungs. Other cases can be caused by the SLC39A13 gene. Those with variations in this gene have protuberant eyes, wrinkled palms of the hands, tapering fingers, and distal joint hypermobility.
It was soon found that plutonium was being excreted at a very slow rate, accumulating in test subjects involved in early human experimentation. This led to severe health consequences for the patients involved. From April 10, 1945, to July 18, 1947, eighteen people were injected with plutonium as part of the Manhattan Project. Doses administered ranged from 0.095 to 5.9 microcuries (μCi). Albert Stevens, after a (mistaken) terminal cancer diagnosis which seemed to include many organs, was injected in 1945 with plutonium without his informed consent. He was referred to as patient CAL-1 and the plutonium consisted of 3.5 μCi 238Pu and 0.046 μCi 239Pu, giving him an initial body burden of 3.546 μCi (131 kBq) total activity. The fact that he had the highly radioactive plutonium-238 (produced in the 60-inch cyclotron at the Crocker Laboratory by deuteron bombardment of natural uranium) contributed heavily to his long-term dose. Had all of the plutonium given to Stevens been the long-lived 239Pu as used in similar experiments of the time, Stevens's lifetime dose would have been significantly smaller. The short half-life of 87.7 years of 238Pu means that a large amount of it decayed during its time inside his body, especially when compared to the 24,100 year half-life of 239Pu. After his initial "cancer" surgery removed many non-cancerous "tumors", Stevens survived for about 20 years after his experimental dose of plutonium before succumbing to heart disease; he had received the highest known accumulated radiation dose of any human patient.
"Neats" hope that intelligent behaviour is described using simple, elegant principles (such as logic or optimisation). "Scruffies" expect that it necessarily requires solving a large number of unrelated problems. Neats defend their programs with theoretical rigour, scruffies rely mainly on incremental testing to see if they work. This issue was actively discussed in the 1970s and 1980s. The rise of deep learning may represent a shift toward the scruffies.
On 19 November, the communists—meeting in Mukachevo—issued a resolution requesting separation of Subcarpathian Ruthenia from Czechoslovakia and incorporation into the Ukrainian Soviet Socialist Republic. On 26 November, the Congress of National Committees unanimously accepted the resolution of the communists. The congress elected the National Council and instructed that a delegation be sent to Moscow to discuss union. The Czechoslovak delegation was asked to leave Subcarpathian Ruthenia. Negotiations between the Czechoslovak government and Moscow ensued. Both Czech and Slovak communists encouraged Beneš to cede Subcarpathian Ruthenia. The Soviet Union agreed to postpone annexation until the postwar period to avoid compromising Beneš's policy based on the pre-Munich frontiers. The treaty ceding Carpathian Ruthenia to the Soviet Union was signed in June 1945. Czechs and Slovaks living in Subcarpathian Ruthenia and Ruthenians (Rusyns) living in Czechoslovakia were given the choice of Czechoslovak or Soviet citizenship.

British Post Office scandal: Secretary of State for Justice Alex Chalk tells Parliament the UK government is giving "serious consideration" to introducing legislation to quash the convictions of the 700 or so sub post masters who were prosecuted as a result of the Horizon IT scandal. Former Post Office chief executive Paula Vennells announces that she will hand back her CBE after more than a million people signed a petition calling for her to do so. Liberal Democrat leader Ed Davey, who was Post Office minister during the scandal, comes under pressure to return his knighthood. 10 January British Post Office scandal: Prime Minister Rishi Sunak announces that emergency legislation will be brought through Parliament to "swiftly exonerate and compensate victims" of the Post Office scandal in England and Wales. First Minister of Scotland Humza Yousaf confirms those in Scotland convicted because of the scandal will also be cleared, and that he will work with the UK government to bring this about. Baroness Heather Hallett, chair of the UK COVID-19 Inquiry, confirms the inquiry will postpone the start of hearing evidence about the development of a vaccine as more time is needed to prepare for a separate investigation into the impact of COVID-19 on the NHS. Consequently, the vaccine evidence, which was due to begin being heard in Summer 2024 may not begin until after the next general election. Nausea: ~ 11%; 1% severe nausea and 2% discontinued treatment due to nausea. Diarrhea: ~ 8%; 2% severe diarrhea and 1% of patients discontinued treatment due to diarrhea. Less common adverse reactions (<1%): fecal incontinence, blood potassium decreased. For subjects with irritable bowel syndrome with constipation:
Glutaredoxin 2 (GLRX2) is an enzyme that in humans, is encoded by the GLRX2 gene. GLRX2, also known as GRX2, is a glutaredoxin family protein and a thiol-disulfide oxidoreductase that maintains cellular thiol homeostasis. This gene consists of four exons and three introns, spanned 10 kilobase pairs, and localized to chromosome 1q31.2–31.3. Alternative splicing of GLRX2 leads to three isoforms of Grx2. One isoform, Grx2a, localizes to the mitochondria, is ubiquitously expressed in tissues (e.g. heart, skeletal muscle, kidney, and liver), regulates mitochondrial redox homeostasis, and protects cells against oxidative stress. Isoforms Grx2b and Grx2c, both localized to the nucleus and cytosol, are expressed only in testes and cancer cell lines and facilitate cellular differentiation and transformation, potentially inducing tumor progression.

=== Coloring in consumer products === Consumer product uses have included incorporation into dental porcelain, used for false teeth to simulate the fluorescence of natural teeth, and uranium-bearing reagents used in chemistry laboratories (e.g. uranyl acetate, used in analytical chemistry and as a stain in electron microscopy). Uranium (both depleted uranium and natural uranium) was widely used as a coloring matter for porcelain and glass in the 19th and early-to-mid-20th century. The practice was largely discontinued in the late 20th century. In 1999, concentrations of 10% depleted uranium were being used in "jaune no.17" a yellow enamel powder that was being produced in France by Cristallerie de Saint-Paul, a manufacturer of enamel pigments. The depleted uranium used in the powder was sold by Cogéma's Pierrelatte facility. In February 2000, Cogema discontinued the sale of depleted uranium to producers of enamel and glass.

Trials differ in patient population, dose schedule, background treatment, and the endpoints used to judge success. Many are small and single-center, so random variation can dominate the reported effects.
Authorization is limited to a small number of countries and covers specific indications such as chronic hepatitis B and vaccine adjuvant use. Availability and labelling differ by jurisdiction.
Most reviews describe it as an immunomodulatory agent with an uncertain clinical effect. They generally call for larger, better-controlled trials before firm conclusions are drawn.
The lyophilized powder is kept refrigerated at 2 to 8 degrees Celsius and protected from light. Reconstituted solutions should be used promptly. Freezing and thawing repeatedly is avoided.
