The peptide was identified during work in the 1970s on thymosin fraction 5, a partially purified extract of calf thymus. Investigators separated that mixture and characterized individual components, one of which they named thymosin alpha-1. The same compound later received the international nonproprietary name thymalfasin. Commercial material is produced by solid-phase peptide synthesis rather than by extraction, so synthetic and natural forms share an identical sequence. Naming conventions vary across the literature, and readers should distinguish the alpha-1 peptide from other thymosins that have unrelated sequences and functions.
Thymosin beta-4 is a separate 43-residue peptide that binds actin and participates in cell migration; it shares no sequence similarity with thymosin alpha-1 despite the common family name. Other preparative materials, such as thymosin fraction 5 and thymopoietin, contain distinct mixtures or peptides. The shared thymosin label reflects the tissue of origin used in early purification, not a common structural core. Treating these molecules as interchangeable is a frequent source of confusion in laboratory reports and in popular summaries alike.
The peptide was described in the 1970s as a component of thymic extracts, and early research focused on restoring immune function in immunodeficiency states. A synthetic version entered clinical development in the 1980s and is approved as a drug in several countries for conditions such as chronic hepatitis B and certain immunodeficiencies. Approval status varies widely by jurisdiction, and in the United States it is not an approved therapeutic. Regulatory and clinical positions differ, so statements about efficacy should be tied to specific indications and studies.
Thymosin alpha-1 is a synthetic peptide of 28 amino acids, corresponding to the N-terminal fragment of prothymosin alpha. Its sequence begins with acetylation at the N-terminus, a modification that affects stability and receptor interaction. The peptide is acidic, with a calculated isoelectric point near 4.2, and carries no disulfide bonds, so its secondary structure is largely flexible in solution. Molecular mass is approximately 3108 daltons. The native form was first isolated from bovine thymus tissue, while pharmaceutical material is produced by solid-phase peptide synthesis.
| Property | Value | Notes |

| Solubility | Freely soluble in water | Also soluble in aqueous buffers |
| Storage temperature | 2 to 8 °C | Protect from light and moisture |
| Common synonyms | Thymalfasin; Tα1 | Same peptide sequence |
Whether the free 28-residue peptide circulates in human tissue remains debated. The best-documented human source is prothymosin alpha, a larger acidic protein that carries the sequence at its N-terminus. Reports of measurable peptide levels in serum and lymphoid tissue exist, yet some of that signal may come from cross-reacting fragments or from the parent protein. Most reviews therefore treat prothymosin alpha as the established human molecule and describe independent circulation of the small peptide as an unresolved question.
Immunological studies connect the peptide to multiple parts of the immune response. It has been reported to engage Toll-like receptor signaling, to promote dendritic cell maturation, and to influence the balance of T helper cell subsets. Changes in natural killer cell activity and in cytokine release appear in cell culture and animal models. These observations describe broad immunomodulatory behavior rather than a single defined receptor target, and the primary molecular interaction has not been settled.
Clinical research has examined the peptide in chronic hepatitis B and C, as a vaccine adjuvant, and in sepsis and oncology settings. Findings across trials are mixed; some report changes in selected immune markers, while others find no clear clinical benefit. Many studies are small and define outcomes differently, which limits comparison. Regulatory approval is confined to a few countries, and the compound is not an approved drug in the United States or most of Europe.
Overall evidence quality varies considerably. A large share of published reports come from single centers, rely on surrogate immunological markers, or lack adequate control groups. Systematic reviews have highlighted this heterogeneity as a barrier to pooling results. Open questions include which patients, if any, might benefit, what treatment duration is appropriate, and whether any effect is independent of standard care. The peptide is often described as an immune modulator rather than a therapy for one disease, which complicates confirmatory trial design.
Thymosin alpha 1 was identified in 1977 as a component of thymosin fraction 5, a heterogeneous preparation used in early studies of thymic function. Investigators purified the active material and determined its amino acid sequence, which enabled chemical synthesis. Work in the following decades concentrated on T-cell maturation and immune reconstitution in animals and small human cohorts. Early preparations varied in composition, so results from that period are difficult to compare with studies using defined synthetic peptide.
=== Post-processing === In the post-processing phase, printed food may require additional steps before consumption. This includes processing activities such as baking, frying, cleaning, etc. This phase can be one of the most critical to 3D printed food, as the printed food needs to be safe for consumption. An additional concern in post processing is the deformation of the printed food due to the strain of these additional processes. Current methods involve trial and error. That is, combining food additives with the materials/ingredients to improve the integrity of complex structures and to ensure the printed structure retains its shape. Additives such as transglutaminase and hydrocolloids have been added to ingredients in order to help retain the printed shape while printing and after cooking. Additionally, recent research has produced a visual simulation for baking breads, cookies, pancakes and similar materials that consist of dough or batter (mixtures of water, flour, eggs, fat, sugar and leavening agents). By adjusting certain parameters in the simulation, it shows the realistic effect that baking will have on the food. With further research and development, a visual simulation of 3D printed foods being cooked could predict what is vulnerable to deformation.
In Japan in 675, the Emperor Tenmu prohibited the killing and the eating of meat during the busy farming period between April and September but excluded the eating of wild birds and wild animals. These bans and several others that followed over the centuries were overturned in the nineteenth century during the Meiji Restoration. In China, during the Song dynasty, Buddhist cuisine became popular enough that vegetarian restaurants appeared where chefs used ingredients such as beans, gluten, root vegetables and mushrooms to create meat analogues including pork, fowl, eggs and crab roe and many meat substitutes used even today such as tofu, seitan and konjac originate in Chinese Buddhist cuisine.
== History == While studying real estate at Ohio State University, Korean American Charley Shin worked in his mother's Japanese/Korean restaurant in Columbus, Ohio. While taking a trip to New York City, the Shins took a detour to Philadelphia, resulting in Charley's discovery of the Philly cheesesteak. Charley convinced his mother to take a break, sell the restaurant, and invest her life savings of $48,000 in "Charley's," a 450-square-foot (42 m2) restaurant across the street from Ohio State's campus.
=== Chimeric and humanized === To reduce murine antibody immunogenicity (attacks by the immune system against the antibody), murine molecules were engineered to remove immunogenic content and to increase immunologic efficiency. This was initially achieved by the production of chimeric (suffix -ximab) and humanized antibodies (suffix -zumab). Chimeric antibodies are composed of murine variable regions fused onto human constant regions. Taking human gene sequences from the kappa light chain and the IgG1 heavy chain results in antibodies that are approximately 65-75% human. This reduces immunogenicity, and thus increases serum half-life as the chimeric or humanized antibody is less likely to be recognized as foreign by the bodies immune system. Humanised antibodies are produced by grafting murine hypervariable regions on amino acid domains into human antibodies. This results in a molecule of approximately 95% human origin. Humanised antibodies bind antigen much more weakly than the parent murine monoclonal antibody, with reported decreases in affinity of up to several hundredfold. Increases in antibody-antigen binding strength have been achieved by introducing mutations into the complementarity determining regions (CDR), using techniques such as chain-shuffling, randomization of complementarity-determining regions and antibodies with mutations within the variable regions induced by error-prone PCR, E. coli mutator strains and site-specific mutagenesis.
Another important application of nitrile rubber is protective gloves, which are frequently used in healthcare instead of latex clothing gloves, as the latter often cause latex allergies. Such gloves are also commonly used when handling hazardous chemicals, including organic solvents. Another important polymer is the terpolymer of acrylonitrile, butadiene, and styrene (acrylonitrile-butadiene-styrene copolymer). This material is widely used for the outer housings of electronic devices (computers, monitors, and keyboards). Other applications include automotive plastic components (e.g., headlight and mirror housings), refrigerator liners, housings for kitchen appliances, vacuum cleaners, and power tools, as well as suitcases, snack containers, and toys, including Lego. ABS is also produced on the scale of several million tons annually; for example, about 2.7 million tons were manufactured in 1992. Polyamide (nylon) is not a nitrile polymer; however, a key intermediate in its production is adiponitrile. Adiponitrile is obtained by hydrocyanation of butadiene or by dimerization of acrylonitrile and is converted by catalytic hydrogenation into hexamethylenediamine, one of the monomers used to produce nylon. The second monomer, adipic acid, is produced by oxidation of cyclohexane. Acetone cyanohydrin is an important intermediate in the production of methyl methacrylate, which in turn is used to manufacture polymethyl methacrylate.

=== Food preservative === In foods, TBHQ is used as an antioxidant preservative for unsaturated vegetable oils and many edible animal fats. It does not cause discoloration even in the presence of iron, and does not change flavor or odor of the material to which it is added. It can be combined with other preservatives such as butylated hydroxyanisole (BHA). As a food additive, its E number is E319. It is added to a wide range of foods. Its primary advantage is extending storage life.
Holick helped develop the first clinical assays for 25-hydroxyvitamin D and 1,25-dihydroxyvitamin D, determined how vitamin D3 is made in the skin from sun exposure, and established how season, time of day, skin pigmentation, sunscreen use, and latitude influenced this vital cutaneous process. He established that the skin was not only the organ responsible for making vitamin D3 but was also a target tissue for its active form, 1,25-dihydroxyvitamin D3. He determined the extremely inhibitory effects of 1,25-dihydroxyvitamin D3 on keratinocyte proliferation and the promoting effects on differentiation, and translated these seminal observations by demonstrating that the topical application of 1,25-dihydroxyvitamin D3 and several of its analogs were effective for the treatment of psoriasis. He demonstrated that macrophages and prostate cells have the enzymatic machinery to produce 1,25-dihydroxyvitamin D3, and established that the extrarenal production of 1,25-dihydroxyvitamin D3 may play a crucial role not only in cancer prevention but also in regulating the immune system. He developed a vitamin D absorption test and demonstrated that vitamin D was bioavailable in orange juice, leading to fortification of juice products in the United States. He also used the test to demonstrate the major cause of vitamin D deficiency in obesity is sequestration of vitamin D in the fat. He helped perform dose escalation studies establishing how much vitamin D is required to maintain blood levels of 25-hydroxyvitamin D in the sufficient range for adults.
== Medical uses == Trimipramine's primary use in medicine is in the treatment of major depressive disorder, especially where sedation is helpful due to its prominent sedative effects. The drug is also an effective anxiolytic, and can be used in the treatment of anxiety. In addition to depression and anxiety, trimipramine is effective in the treatment of insomnia, and unlike most other hypnotics, does not alter the normal sleep architecture. In particular, it does not suppress REM sleep, and dreams are said to "brighten" during treatment. Trimipramine also has some weak antipsychotic effects with a profile of activity described as similar to that of clozapine, and may be useful in the treatment of psychotic symptoms, such as in delusional depression, schizoaffective disorder or schizophrenia. A major systematic review and network meta-analysis of medications for the treatment of insomnia published in 2022 found that trimipramine had an effect size (standardized mean difference (SMD)) against placebo for treatment of insomnia at 4 weeks of 0.55 (95% CITooltip confidence interval –0.11 to 1.21). The certainty of evidence was rated as very low, and no data were available for longer-term treatment (3 months). For comparison, the other sedating antihistamines assessed, doxepin and doxylamine, had effect sizes (SMD) at 4 weeks of 0.30 (95% CI –0.05 to 0.64) (very low certainty evidence) and 0.47 (95% CI 0.06 to 0.89) (moderate certainty evidence), respectively. The effective dosage of trimipramine in depression is 150 to 300 mg/day.
== With herbal medicines == Herb-drug interactions are drug interactions that occur between herbal medicines and conventional drugs. These types of interactions may be more common than drug-drug interactions because herbal medicines often contain multiple pharmacologically active ingredients, while conventional drugs typically contain only one. Some such interactions are clinically significant, although most herbal remedies are not associated with drug interactions causing serious consequences. Most catalogued herb-drug interactions are moderate in severity. The most commonly implicated conventional drugs in herb-drug interactions are warfarin, insulin, aspirin, digoxin, and ticlopidine, due to their narrow therapeutic indices. The most commonly implicated herbs involved in such interactions are those containing St. John's Wort, magnesium, calcium, iron, or ginkgo.

It is a 28-residue synthetic peptide studied as an immune-modulating agent and approved as a drug in some countries. The sequence matches a naturally occurring fragment isolated from thymus tissue. It is not a hormone in the endocrine sense.
The name traces back to thymosin fraction 5, a crude thymus extract examined in the 1970s. Individual peptides in that mixture were labeled with Greek letters, and alpha-1 was one of them. The international nonproprietary name thymalfasin was assigned later.
No. Thymosin beta-4 contains 43 residues and binds actin, while thymosin alpha-1 contains 28 residues and acts on immune cells. The two sit in a historical naming group but share no sequence similarity, and they are not substitutes for one another.
It corresponds to a fragment of the larger protein prothymosin alpha, which is present in many tissues. The isolated 28-amino-acid peptide was originally obtained from thymus preparations, and the pharmaceutical product is synthesized rather than extracted. The term therefore describes both a natural fragment and a manufactured drug substance.
